Tyra emphasizes that melatonin is often misunderstood as solely a sleep aid, but it is in fact one of the body's most potent antioxidants and free radical scavengers. Melatonin protects cells from oxidative stress and inflammation, supports insulin sensitivity, and promotes mitochondrial health—the energy-producing organelles within cells. These functions contribute to overall health and energy levels, particularly relevant for those experiencing fatigue.
Melatonin also plays a role in reproductive health by regulating the timing and release of sex hormones. It is synthesized in ovarian follicular cells, oocytes (female egg cells), and the placenta. Thanks to its antioxidant properties, melatonin helps preserve oocyte quality, making it a potentially important supplement for individuals facing infertility challenges.
Research highlighted in the episode shows that melatonin receptor gene polymorphisms are significantly associated with PCOS. Melatonin supplementation has been found to improve menstrual irregularities and reduce hyperandrogenism (excess androgens) in women with PCOS. One study cited demonstrated that six months of melatonin treatment decreased androgen levels, increased follicle-stimulating hormone (FSH), and lowered anti-Müllerian hormone (AMH) levels, indicating improved ovarian function.
Melatonin supports immune function and exhibits antiviral activity at the cellular level. Tyra notes that melatonin was included in early COVID-19 treatment kits due to these properties. Additionally, melatonin may slow the progression of neurodegenerative diseases such as Alzheimer's by aiding the glymphatic system, a recently discovered brain waste clearance network that removes toxins during sleep.
Melatonin also helps regulate circadian rhythms, which are often disrupted in cancer patients, contributing to altered cortisol patterns and overall hormonal imbalance.
While melatonin is produced in the pineal gland in the brain, Tyra explains that up to 400 times more melatonin is synthesized in the gut mucosa by enterochromaffin cells. Unlike pineal melatonin, gut-derived melatonin is not regulated by light and dark cycles but is influenced by food intake and digestion.
Dietary sources of melatonin include animal foods such as eggs and fish (highest melatonin content), as well as plant foods like nuts, mushrooms, cereals, and germinated legumes or seeds. However, melatonin content varies widely even within the same food types, so diet alone is insufficient to maintain optimal levels.
Melatonin supports gut health by increasing the tone of the lower esophageal sphincter (LES), which helps prevent acid reflux and heartburn by preventing stomach contents from flowing back into the esophagus. It also promotes regeneration of epithelial cells lining the gastrointestinal tract, which naturally atrophies with age.
Melatonin possesses gastroprotective properties, including regulation of gut motility, intestinal permeability, and mucosal healing. It has been shown to aid healing of gastric ulcers caused by Helicobacter pylori infection and non-steroidal anti-inflammatory drugs (NSAIDs), with clinical trials demonstrating comparable effects to proton pump inhibitors (PPIs) like omeprazole (Prilosec).
Clinical trials have also used melatonin at doses of 3 mg per day to successfully improve symptoms of irritable bowel syndrome (IBS), including both constipation-predominant and diarrhea-predominant types. Tyra shares her clinical experience using melatonin to support patients with heartburn, IBS, gastritis, stomatitis, esophagitis, peptic ulcers, and to assist in weaning off PPIs.
Tyra discusses melatonin’s promising role in cancer treatment support, particularly its ability to inhibit the Warburg effect—a metabolic shift where cancer cells preferentially use glycolysis (sugar metabolism) for energy, fueling rapid tumor growth. Melatonin supplementation has been shown in human clinical trials to reduce tumor cell proliferation, metastasis, and drug resistance, and to improve chemotherapy response rates in some lung cancer patients.
Additionally, melatonin helps restore disrupted circadian rhythms and cortisol patterns commonly observed in breast and colon cancer patients, supporting overall hormonal balance and potentially improving treatment outcomes.
Melatonin levels naturally decline starting in the 20s, similar to other hormones. This decline is exacerbated by lifestyle factors such as exposure to artificial and blue light at night, stress, poor diet, gut dysbiosis, environmental toxins, and certain medications. Symptoms of low melatonin include sleep disturbances, oxidative stress, inflammation, menstrual irregularities, infertility, anxiety, migraines, immune dysfunction, and increased risk for chronic diseases including cancer, heart disease, and Alzheimer’s disease.

Tyra cautions against lifestyles that involve waking in darkness, spending the day indoors under artificial light, and exposure to screens emitting blue light in the evening, describing this pattern as a recipe for melatonin deficiency and hormonal disruption.
Tyra prefers plant-based melatonin supplements (phytomelatonin) such as Herbatonin, which she uses personally and recommends to clients. Research indicates that phytomelatonin is 470 to 957 percent more effective as an antioxidant and up to 646 percent more effective as an anti-inflammatory compared to synthetic melatonin. This enhanced effect may be due to additional antioxidant compounds present in plant-based sources, such as chlorophyll, beta-carotene, and lutein.
Herbatonin is available in two doses: a physiological dose of 0.3 mg, ideal for long-term use, and a higher 3 mg dose, useful for gastrointestinal conditions. Tyra recommends taking 0.3 mg about 30 minutes before bedtime for general support.

Side effects of melatonin supplementation can include grogginess, nausea, vivid dreams, and irritability. Tyra explains that grogginess may be related to brain detoxification via the glymphatic system, which melatonin supports by facilitating toxin clearance during sleep. To mitigate grogginess, she suggests taking melatonin with healthy fats (e.g., almond butter), using binders to remove toxins, or supplementing with the amino acid glycine, which also promotes sleep.
Tyra dispels the myth that melatonin supplementation stops the body’s own production, citing research showing that even high doses (up to 50 mg daily) do not suppress endogenous melatonin. However, long-term use of high doses may reduce melatonin receptor sensitivity, so physiological doses are preferred for ongoing use.
Tyra advises caution when considering melatonin supplementation for children, especially those approaching puberty, due to melatonin’s influence on reproductive hormones. She recommends prioritizing lifestyle strategies to support natural melatonin production before considering supplementation in this population.
